| Beschreibung | The Research Center Pharmaceutical Engineering GmbH (RCPE) is a Research Center in the field of Pharmaceutical Engineering. Our goal is to establish technology to transform current product and processes development approaches in the pharmaceutical and diagnostics industries into a science-based endeavor together with our partners. PAID DIPLOMA OR MASTER THESIS Conformational changes of pharmaceutical proteins in the frozen state Ref.Nr. DA24 SCOPE OF THE PROJECT: Long term storage of liquid protein pharmaceuticals is very often facilitated by freezing. Industrial scale freezing entails side effects such as freeze concentration, pH shifts, and surface effects from water crystallization, resulting in irreversible freeze denaturation. A 300 mL lab-scale freeze container was constructed as a model for a 300 L commercial freezing container by Zeta Biopharma, Austria. Freeze damage to several protein pharmaceuticals shall be examinded for freezing and thawing in the lab scale container. Especially changes in protein conformation during freezing and thawing will be analyzed using hydrogen/deuterium exchange mass spectrometry (HDX-MS, mainly operated at the Medical University Graz). Also biophysical characterization and activity assays are expected to provide information about freezing performance. Results will be cross-validated with computational fluid dynamics simulations. The main part of the work will be performed at the Institute for Biotechnology and Biochemical Engineering, University of Technolgy, Petersgasse 12, A-8010 Graz YOUR DUTIES AND RESPONSIBILITIES: - Operation of a lab-scale freeze container, preparation and analysis of HDX-MS experiments, biophysical characterization of freeze/thaw behaviour of pharmaceutical proteins - Background in biochemistry, biotechnology or a related field - Motivation for interdisciplinary work at the interface between biochemistry, process engineering and computational sciences. - Interest in basic and applied research in close collaboration with pharmaceutical and biotech industry START: may start work immediately We offer - A multidisciplinary and dynamic research environment in the area of pharmaceutical technology, - Mentoring and on-the-job training by our qualified staff, and - An adequate payment as well as the chance for personal and technical development in the area of pharmaceutical engineering.
